Recobrado de Pseudomonas syringae pv. tabaci a partir de semillas de tabaco infectadas

Abstract
Tobacco seeds are an important way for disseminating Pseudomonas syringae pv. tabaci. A procedure for recovering the bacteria from inoculated seeds was tried, which consisted in the extraction of bacteria in PBS, continued with centrifuging the supernatant, sowing in a King B medium and later the inoculation of recovered liquid by infiltration in tobacco leaves. To determine the method sensitivity, work samples were formed with different amounts of inoculated and healthy seeds to complement one gram. Only an average of 196.6 bacteria colonies/dish (107 cfu/mL) was recovered from the sample of inoculated seeds. High levels of the pathogen, within the range of 106 to 105 cfu/mL were detected in treatments contained infected seeds from 0.5 to 0.1 g. Bacterial concentration of 107 cfu/mL caused a necrosis surrounded by a chlorotic halo on tobacco leaves over a period of 24 hours, concentrations in the range of 104 cfu/mL produced chlorosis and infection expansion from 3-4 days. Injuries corresponding to lower populations (103-101) appeared between 10-15 days in the form of small humid spots and light chlorosis. It was possible to detect the presence of Pseudomonas syringae pv. Tabaci in the seeds, by sowing in the culture medium with concentrations up to 103 cfu/mL and with infiltration of tobacco leaves, it was not possible to isolate colonies of the bacteria in lower dilutions (10–2-10–1).
